The rapid cassette test for COVID-19 antigens is a membrane immunochromatographic test that uses highly sensitive monoclonal antibodies to detect the nucleocapsid protein from SARS-CoV-2 in the nasopharyngeal secretion (NP) sample. The test strip is composed of the following parts: the sample collection pad, the reagent pad, the reagent membrane and the absorbent pad. The reagent buffer contains colloidal gold conjugated with the antibodies
monoclonals against the nucleocapsid protein of SARS-CoV-2; the reactive membrane contains the secondary antibodies against the nucleocapsid protein of SARS-CoV-2.

The whole band is fixed inside a plastic device. When the sample is added to the sample well, the dried conjugates on the reagent support dissolve and migrate with the sample. If SARS-CoV-2 antigens are present in the sample, a complex formed between the anti-SARS-2 conjugate and the virus will be captured by the specific anti-SARS-2 monoclonal antibodies present in the test line (T) region. The absence of the T line suggests a negative result. For procedural control purposes, a red line will always appear in the control line region (C), indicating that an adequate volume of sample has been added and membrane impregnation has occurred.
